Your disk space can do up and down...it doesn't honestly matter but the genral defragmenting process will help your PC.

You should be fine. The real problem is if after it's done you really are missing some files you knew you had.
Also, it's best to not have any programs running or be using the computer while it is defragmenting. It will only make it take longer and won't be as efficient.

Run CCleaner or the disk cleanup tool before the defrag. That will help delete unnecessary files and create free space. The defragmenter only puts fragmented files back together contiguously, helping to improve file access. If you are having trouble with the built in defragger, download a third party tool. Some of the new completely automatic defraggers even run the task with very less free space.
